Human IRAK4 Xpress Clones express IRAK4, a 52 kDa serine/threonine kinase critical in IL-1R and TLR signaling pathways, activating transcription factors like NF-κB and AP-1. The protein contains an N-terminal death domain (DD) and a central kinase domain (KD) but lacks the extended C-terminal region present in some IRAKs. The KK213AA mutation in the ATP-binding pocket makes IRAK4 catalytically inactive, significantly reducing IL-1-induced NF-κB activation, unlike kinase-deficient IRAK1. This highlights the essential role of IRAK4’s kinase activity in immune signaling. Expression is highest in kidney and liver, with low levels in other tissues. - 20 ug
